The SureGuide design wizard allows you to use a custom genome for selection of gRNA sequences. On the My Genomes dialog box, you can manage the existing custom genomes and upload the sequence file for new a custom genome. Once a custom genome has been uploaded, it is available in your workgroup as a species selection when creating SureGuide designs.
To open the My Genomes dialog box: From the Home screen, cick Find Designs > SureGuide. Select the My Designs tab. On the left side of the screen, on the Species panel, click Manage >My Genomes. Or, from the SureGuide design wizard, click Manage Genomes at the Define Design step.
To upload a custom genome:
Open the My Genomes dialog box.
Click Add
Genome.
Your internet browser opens the Genome File Upload page opens in a
new tab or new window.
NOTE Before continuing, make sure
your genome file is in the proper format. Read the guidelines on the
right side of the Genome File Upload page for information on the genome
file requirements. To view a sample genome file, click the Sample-File
link next to Template.
In the Species drop-down list, select the
species of the custom genome.
If the appropriate species is not included in the list, select Custom.
In the Build Version field, type the build version of the custom genome.
(Optional) If desired, type a description into the Description field.
Click Choose
File and Upload.
The Open dialog box opens.
Browse to the genome file. Select the file
and click Open.
A progress bar opens showing the progress of the upload.
If the file uploads successfully, the SureDesign system sends you an e-mail when the progress bar reaches 100%. The genome is now available for selection as the species designation when creating a new SureGuide design. Close the Genome File Upload page. In the My Genomes dialog box, click Refresh to see the new genome listed in the table.
If the file fails to upload, the Genome File Upload page displays an error message. Check the format of the genome file for errors. The Sample-File link on the Genome File Upload page opens a template of a correctly formatted genome file.
To delete a custom genome:
Open the My Genomes dialog box.
In the table on the dialog box, locate the custom genome that you want to delete. In the Action column, click Delete.
In the message box that opens, click OK to continue.
The custom genome is removed from your workgroup.
