When you create a SurePrint OLS design, you upload the oligo sequences from a file.
Follow these requirements when preparing an oligo library sequences upload file:
· The file must be a tab-delimited text file (*.txt or *.tdt), a comma-separated values file (*.csv), or a list file (*.list). The file must be saved within a compressed folder (*.zip).
· If your file includes comments, the comment lines must start with a "#" symbol.
· The file must be in a 1-column or 3-column format, with each column and column header completed as described in the tables below.
The column of a 1-column oligo sequences upload file for SurePrint OLS designs is described in the table below.
Column # |
Column header |
Description of column content |
1 |
Sequence |
This column contains the complete sequence of the oligo in 5' to 3' orientation, with one oligo sequence per line. The sequence can contain only A, T, G, and C characters. Each sequence must be 30-230 nucleotides long. The same oligo sequences can be listed more than once in the file. |
The columns of a 3-column oligo sequences upload file for SurePrint OLS designs are described in the table below.
Column # |
Column header |
Description of column content |
1 |
SequenceName |
The identifier for the oligo sequence. |
2 |
Sequence |
This column contains the complete sequence of the oligo in 5' to 3' orientation, with one oligo sequence per line. The sequence can contain only A, T, G, and C characters. Each sequence must be 30-230 nucleotides long. |
3 |
Replication |
The number in this column indicates the number of times that the oligo sequence is replicated within the library. |
