Use the SurePrint OLS design wizard to create custom oligo libraries with sequences that you upload directly into SureDesign. You provide a file containing the oligo information, including sequences, and submit the file to SureDesign as an upload job. SureDesign uploads the oligos and saves them to the SurePrint OLS design.
The wizard takes you through the steps of the design creation process, which are described in detail below. The end-result is a set of design files that includes the oligo sequences and related information. When you place an order for a SurePrint OLS design, you can use the DNA oligos in a variety of downstream applications.
NOTE Prepare your oligo sequences file before you begin the SurePrint OLS wizard. The maximum number of sequences permitted per file is 244,000 (including replicates). The file must be a tab-delimited (saved with file extension txt, tdt, csv, or list) in a 1-column or 3-column format. For upload purposes, create a compressed (*.zip format) version of the file. To open the SurePrint OLS design wizard:
· At the top of the screen, click Create Designs > SurePrint OLS.
The wizard window opens to Step 1.
In this step, complete the fields described below to define the design.
Design Name
Type a name for your design into the field. Alphanumeric characters, hyphens, underscores, and spaces are permitted. The name must be unique within your workgroup.
Specify the folder in which you want to save this design. The default selection is the top-level folder for your workgroup.
To change the selection, click Select to open the Select Folder dialog box, and mark the folder in which you want to save the new design. This dialog box lists the available folders within your workgroup.
Description
(Optional) Type a description of the design. The description can be up to 2000 characters. Use only alphanumeric characters (a-z, A-Z, and 0-9) and the following special characters: _, -, $, #, @.
Keywords
(Optional) Type one or more keywords for the design, separated with a comma or space. You can type up to 2000 characters in this field. Use only alphanumeric characters (a-z, A-Z, and 0-9), commas, dashes, and underscores in your keywords. You can later use these keywords to search for the design.
Click Next to advance to Step 2.
In this step, click Browse to select the oligo sequences upload file (in *.zip format). Alternatively, if you know the file path for the file, you can type it directly into the File Name field.
To submit the selected file for upload:
When you are ready to upload the file, click
Submit.
A message box opens indicating the e-mail address where Agilent will
contact you when SureDesign has finished uploading your oligo sequences.
If desired, you can enter additional e-mail addresses into the provided
field in the message box.
Click OK
in the message box to submit the file.
SureDesign begins the process of uploading your oligo sequences. The
program first scans the upload file for formatting errors. If it detects
a formatting error, you are returned to the Upload Sequences step
of the wizard. If it does not detect any formatting errors, SureDesign
uploads the oligo library into your SureDesign account and the wizard
opens to Step 3: Review.
In this step the oligo upload job is processing.
The wizard opens to Step 4: Finalize.
This step allows you to view statistics on the uploaded oligo sequences before finalizing the design.
When ready, click Finalize Design. The wizard opens to Step 5: Design Complete.
After clicking Finalize Design, the wizard window updates to the Design Complete step.
Use the action buttons at the bottom of the Finalize Design window to take further action on the design:
· Click Mark as Favorite to add the design to your list of favorites. The design will appear in the Designs: Recent and Favorites dashboard on the Home screen.
· Click Download to download one or more design files, including a formatted PDF report that summarizes key information on the design.
· Click Order to place an order or request a quote for the design.
