Overview of working with mutagenesis oligo sets |

A mutagenesis oligo set is a group of DNA oligonucleotides that are created from the application of a mutational strategy to a mutational region in a given target DNA sequence. A mutagenesis library contains one or more oligo sets. For details, see Learn about mutagenesis libraries.
The figure below illustrates a mutational region in a target DNA sequence, and a mutagenesis oligo set that could be created from it. All oligos in the set are the same length, and are identical in sequence to the corresponding region of the target sequence, except for the necessary base changes to create the requested amino acid changes. The mutational regions in the oligos are flanked on both the 5' and 3' ends with sequences that contain PCR primer annealing sites. If space on the oligos and within the target sequence allows, eArray can optimize both the length and position of the PCR primer annealing sites for the set. These PCR primers are used with the QuikChange Oligo Library Mutagenesis System to amplify this oligo set in the custom mutagenesis library.
You can create mutagenesis oligo sets in two ways:
Use the Create Mutagenesis Oligo Sets wizard – This wizard leads you through the creation of mutagenesis oligo set(s) based on the application of one or more mutational strateg(ies) to defined mutational region(s) on a target DNA sequence that you enter. For details, see Create mutagenesis oligo sets (wizard).
Use the Create Mutagenesis Library from Target Sequence wizard – This wizard leads you through the creation of a mutagenesis library based on a target DNA sequence that you enter. The final result of the wizard is a mutagenesis library, but it also creates mutagenesis oligo sets in your eArray account. See Create library from target sequence (wizard).
After you create a mutagenesis oligo set, it becomes available for you
to include in a custom mutagenesis library. A given oligo set can be included
in any number of mutagenesis libraries.
Mutagenesis oligo sets always have a status of Locked. Once an oligo set is created, in cannot be edited.
The table below summarizes the actions that you can take on mutagenesis oligo sets. To edit an oligo set, it must have a status of Incomplete. All other actions are available without regard to the status of the oligo set. For more information, click the links in the table.
